RNA-sequencing analysis of human RTT autopsy samples from the motor cortex and cerebellum identified genes that are increased or decreased. (Gogliotti et al 2018 J Pharmacol Exp Ther) The decreased genes include those encoding for mGlu and muscarinic receptors, which are also seen in temporal cortex samples.

 

Western blotting. Paralleling mRNA expression, protein expression of mGlu5 receptors in the human RTT autopsy samples is decreased. (Gogliotti et al 2016 Hum Mol Genet). Flow Cytometry. Single nuclei are isolated from Mecp2+/- mouse brain tissue. Flow cytometry analysis is used to characterize two nuclei populations – Mecp2 mutant (mut) or wild-type (WT).
