Yeast media
Yeast YPD Media (per Liter)
10g Yeast extract
20g Bacto Peptone
20g glucose
(for plates add 20g Bacto Agar)
Yeast Minimal (Dropout) Media (per Liter)
20g glucose
2g dropout mix
900ml water
for plates:
100ul of 10N NaOH or 1 NaOH pellet
20g of Agar
After autoclaving add 100ml of 10XYNB and 5ml of 200X Tryptophan
10X YNB
50g Ammonium sulfate
17g Yeast Nitrogen Base
Water to 1L
Filter sterilize and wrap in aluminum foil
Dropout mixes

1.523g/100ml filter sterilize -- You must add tryptophan after autoclaving because it is sensitive to autoclaving
Galactose Media
For galactose media 100ml of 20% galactose is substituted for the 20g of glucose in the recipe (per Liter).� Make 20% galactose by filter sterilizing--not autoclaving.
When making galactose plates Mix 20g of Agar and 800ml of water in a flask autoclave; also autoclave 2g of dropout mix with 1 NaOH pellet in 100ml of water. After autoclaving mix the contents of the two flasks. Then add 100 ml of 20% galactose, 100ml of 10X YNB and 5ml of 200x tryptophan. (If you autoclave agar and the dropout mix together in the absence of glucose then the agar turns gray the glucose in the normal minimal media prevents this.)
